She almost certainly knows about the Midnight Fane, and she would be the only person we know of that would even think of setting up an Alter of Desna in it! To access the Desna alter you have to pass a very difficult arcane skill check (I think its 40 DC), if you fail the check you're instead teleported near a portal and will trigger trap.
You need speak to the Hand after you got the key. You get it by doing the Mythic quest which gives you your mythic rank 4.
